Histochemical Methods for Protein-bound Sulphydryl and Disulphide Groups By G. GOMORI

چکیده

Histochemical methods for the localization of protein-bound sulphydryl groups were compared in a critical fashion. As far as localization is concerned, several of the methods published were found to be satisfactory. However, from the point of view of approximate visual quantitation, the results are confusing. The two best methods for the histochemical detection of protein-bound sulphydryl groups are the tetrazolium method and Barrnett and Seligman's dihydroxydinaphthyldisulphide technique.

Distribution of protein sulphydryls and disulphides in fixed mammalian chromosomes, and their relationship to banding.

Fixed mammalian mitotic chromosomes, when stained with a variety of sensitive, sulphydryl-specific fluorochromes directly or following reduction of disulphide groups, show uniform fluorescence. Thus sulphydryl and disulphide groups are uniformly distributed along the length of the chromosomes, and do not show patterns related to chromosome bands. Performance of G- or C-banding procedures oxidiz...

Heat-induced aggregation and sulphydryl/disulphide reaction products of soy protein with different sulphydryl contents.

In this study, soy proteins were reduced with 0.1-10mM dithiothreitol (DTT) to obtain an increasing number of sulphydryl groups (SH) with a similar particle size. Aggregation was promoted by increasing the degree of reduction when heated (100°C, 30min), resulting in larger sized aggregates (from 40 to 70nm) and a higher viscosity of the aggregate dispersion. The disulphide bond (SS) content dec...

Localization and Study of Histochemical Effects

A high capacity for accumulation of Mn was reported for sunflower plants. Localization of excess Mn is therefore of special interest for understanding metal tolerance mechanisms in this species. In this study, structural and histochemical alterations caused by Mn accumulation in leaves were investigated in sunflower (Helianthus annuus L. cv. Azar-ghol) plants grown in nutrient solution. In the ...

The Cytochemistry of the Amoebocytes and Intestinal Epithelium of Venus mercenaria (Lamellibranchiata), with Remarks on a Pigment resembling Ceroid By SUMNER I. ZACKS

The properties of the amoebocytes and intestinal epithelium of Venus mercenaria were studied by a variety of cytochemical procedures designed to demonstrate proteins, enzymes, carbohydrates, and lipids. The cytoplasm of the amoebocytes contains specific granules which are constantly present and which are interpreted as being atypical mitochondria. Identification of their mitochondrial nature re...

Structural mechanism of disulphide bond - mediated redox

The oxidation of cysteine sulphydryl in proteins produces sulphenic acid that can form a reversible disulphide bond with another cysteine. The disulphide bond formation often triggers switches in protein structure and activity, especially when the distance between the two cysteine sulphur atoms is longer than the resulting disulphide bond distance. As an early example for the reversible disulph...
